چکیده

A consistent correlation between elevated esterase activity and methyl parathion resistance among Nebraska western corn rootworm, Diabrotica virgifera virgifera LeConte, populations has previously been documented. Characterization of general esterase activity using naphtholic esters as model substrates indicated that differences between resistant and susceptible strains could be maximized by optimizing assay conditions. The optimal conditions identiÞed here were similar to those reported for other insect species. The majority of general esterase activity was found in the cytosolic fractions of resistant populations, whereas the activity wasmore evenly distributed between cytosolic andmitochondrial/nuclear fractions in the susceptible population. General esterase activity was predominately located in the adult thorax and abdomen. Although there were signiÞcant differences in general esterase activities between resistant and susceptible populations, the differences exhibited in single beetle activity assays did not provide sufÞcient discrimination to identify resistant individuals. In contrast, single larva activity assays provided greater discrimination and could be considered as an alternative to traditional bioassay techniques.

Genetic Differentiation of Western Corn Rootworm Populations (Coleoptera: Chrysomelidae) Relative to Insecticide Resistance

As the single most important pest of field corn, Zea mays L., throughout most of the Corn Belt, the western corn rootworm, Diabrotica virgifera virgifera LeConte (Coleoptera: Chrysomelidae), has undergone repeated selection for resistance to a variety of insecticides that persist widely among Nebraska populations. In this study, we used 11 microsatellite markers to genotype two populations with...

Flight behavior of methyl-parathion-resistant and -susceptible western corn rootworm (Coleoptera: Chrysomelidae) populations from Nebraska.

Relative flight behavior of methyl-parathion-resistant and -susceptible western corn rootworm, Diabrotica virgifera virgifera LeConte populations, was studied as part of a larger effort to characterize the potential impact of insecticide resistance on adult life history traits and to understand the evolution and spread of resistance. A computer interfaced actograph was used to compare flight of...

Cytochrome P450-Mediated N-Demethylation Activity and Induction in Insecticide-Resistant and Susceptible Western Corn Rootworm Populations (Coleoptera: Chrysomelidae)

Elevated cytochrome P450 monooxygenase-based metabolism is associated with carbaryl and methyl parathion resistance in western corn rootworm (Diabrotica virgifera virgifera LeConte) populations in the state of Nebraska. The objectives of this study were to identify potential differences in cytochrome P450 in resistant and susceptible rootworm populations using the model substrate 4-chloro-N-met...
